Summary:This invention describes an assay for human tumor stem cells in explanted cell specimens via a process of observing colony growth on a celled layer system. Described herein are methods and materials for bioassaying the growth of a supporting human tumor stem cell colony, and applications of the bioassay to a variety of neoplasms of differing histopathology. Claims of the invention include techniques by which the bioassay is particularly well suited to the differing growth characteristics and morphologies of tumor stem cell colonies that originate from different types of cancer. Therapeutic applications are included for clinical studies in the effects of anticancer drugs or irradiation on various human tumor stem cell lines.
Abstract:A bioassay method for supporting human tumor stem cell colony growth is disclosed. The method is suitable for culture of a variety of neoplasms of differing histopathology. Tumor stem cell colonies arising from different types of cancer have differing growth characteristics and colony morphology. The present bioassay may be employed in clinical studies of the effects of anticancer drugs or irradiation on human tumor stem cells.
