Summary:Described herein is a method of predicting or detecting agranulocytosis during clozapine therapy. The invention relates to a process by which elevated levels of N-desmethylclozapine may be detected in the blood serum or bone marrow of patients, thereby providing a screening method for measuring the vulnerability of patients on clozapine therapy and their risk of developing agranulocytosis. Further disclosed are methods by which the concentration of N-desmethylclozapine may be established in the blood or bone marrow of the patient and their sensitivity to the drug measured by a process of collecting and assaying heparinized blood for stem cell sensitivity to N-desmethylclozapine.
Abstract:A screening method for detecting the vulnerability of patients on clozapine therapy to developing agranulocytosis which comprises establishing the concentration of N-desmethylclozapine in the blood or bone marrow of said patient. In addition, a method of determining patient sensitivity to N-desmethylclozapine is provided which comprises collecting heparinized blood and assaying it for stem cell sensitivity to N-desmethylclozapine.
