Summary:Described herein are methods of maintaining and expanding mammalian bone marrow cells in a serum-free medium containing a specific mixture of cytokines but essentially free of stromal cells. The invention relates to a process for supporting hematopoietic progenitor cells in a culture medium comprised of the cytokine mast cell growth factor (MGF) wherein the human hematopoietic stem cells are characterized as being CD34+ DR- CD15- and are grown in the presence of at least one cytokine such as interleukin 3 (IL-3) in combination with a granulocyte macrophage colony stimulating factor (GM-CSF). Claims of the invention include an increase in the cell number of the population as high as 20,000 fold when expanded according to this method over a period of 26 days. Therapeutic applications of such cells are included for use in transplantation.
