The invention relates to methods of enriching hematopoietic progenitor cells from body fluids by the use of a cell-trap centrifugation tube containing a gradient solution which is adjusted to a specific density to enrich CD34+ cells from apheresed blood. Further disclosed is the configuration of the apparatus such that a tube allows the desired cell population to be collected by decantation after centrifugation thereby minimizing cell loss. Additional claims of the invention include simplification by density-adjusted cell sorting according to which cell type-specific binding agents are utilized such as antibodies and lectins that are linked to carrier particles thereby imparting a different density to undesired cell populations and allowing the progenitor cells to be separated during centrifugation. Therapeutic applications are included for the use of such donor cell preparations in bone marrow transplantation without the need for invasive procedures such as bone marrow aspiration. | Abstract: | The present invention relates to methods of enriching hematopoietic progenitor cells from body fluids. The tube allows the desired cell population to be collected by decantation after centrifugation to minimize cell loss and maximize efficiency. In addition, the method can be further simplified by density-adjusted cell sorting which uses cell type-specific binding agents such as antibodies and lectins linked to carrier particles to impart a different density to undesired cell populations allowing the progenitor cells to be separated during centrifugation in a more convenient manner.
