Summary:This invention describes a mouse lacking expression of a subunit of the IL-2 receptor in certain cells of the immune system. Central to this concept is a DNA construct comprising a mouse IL-2R? gene fragment spanning the region between the Bgl II site of intron 5 and the Nde I site at the 3' end of exon 7, wherein a neo gene cassette is inserted into the Aat II site of exon 6. Mouse D3 embryonic stem cell lines comprising this DNA construct are then developed. Also disclosed are the uses of such mice, and the methods of such uses.
