Title:Methods, compositions and devices for growing human hematopoietic cells
Inventors:Emerson, Stephen G.; Ann Arbor, MI, USA
Summary:This invention describes a system for culturing human hematopoietic cells via the use of transformed fibroblasts attached to a protein substrate which provides growth factors for the cells. Herein are described methods, materials and techniques for the growth of hematopoietic cells in culture, which includes the use of bioreactors in which diverse cell types are simultaneously cultured in the presence of appropriate levels of nutrients and growth factors which are continuously maintained in the bioreactor while undesirable metabolic products are removed. This simultaneous culturing of multiple cell types is required for the successful reconstruction of hematopoietic tissue ex vivo, as at least one growth factor is provided through excretion by transfected stromal cells, particularly heterologous cells. Further described are the means by which stromal and hematopoietic cells are maintained separately, to allow for early removal of the hematopoietic cells. Methods of proliferation and expansion are also provided.
