Summary:This invention consists of a transgenic mouse which contains in its genome a transgene comprising in operable linkage a plurality of various human genes and associated isotype switching sequences, specific groups of which are located in closer proximity to each other than in the naturally occurring human immunoglobulin locus. The transgenic mouse also contains lymphocytes which undergo productive VDJ rearrangement by recombination between certain switch sequences. The mouse thereby produces a serum containing immunoglobulins of a minimum of 3 human heavy chain isotypes in response to antigenic stimulation.
Abstract:The invention relates to transgenic non-human animals capable of producing heterologous antibodies and methods for producing human sequence antibodies which bind to human antigens with substantial affinity.
