Summary:Described herein is a method of isolating lineage-specific stem cells via the use of a DNA construct comprised of a reporter gene under the control of lineage-specific promoters. The invention relates to the isolation of lineage-specific stem cells in vitro according to a process by which pluripotent embryonic stem cells are transfected with a construct comprised of a regulatory region of a lineage-specific gene that is operably linked to a DNA encoding a reporter protein. Further disclosed are methods by which the pluripotent embryonic stem cells are cultured under conditions which allow for their differentiation into lineage-specific stem cells, and methods for separating those cells which express the reporter protein from other cells in the culture, such that the cells which expresses the reporter protein are identified as isolated lineage-specific stem cells.
Abstract:The present invention provides a method of isolating a lineage specific stem cell in vitro, comprising a) transfecting a pluripotent embryonic stem cell with a construct comprising a regulatory region of a lineage specific gene operably linked to a DNA encoding a reporter protein, b) culturing the pluripotent embryonic stem cell under conditions such that the pluripotent embryonic stem cell differentiates into a lineage specific stem cell and c) separating the cells which express the reporter protein from the other cells in the culture, the cell which expresses the reporter protein being an isolated lineage specific stem cell. A lineage specific stem cell can also be identified utilizing this method.
