Summary:Described herein is a human hematopoietic stem cell composition comprised of stem cells containing exogenous DNA, and methods by which the cells may be assayed for their ability to produce hematopoietic lineages. This invention describes a system for identifying growth factors according to a process by which a sample of Thy-1+ human hematopoietic stem cells is evaluated for its self-regeneration capacity. Further disclosed are methods by which the human hematopoietic stem cells are separated from dedicated cells and maintained by regeneration in an appropriate growth medium, after which they are assayed for their capacity to produce members of each of the hematopoietic lineages. Therapeutic and experimental applications to the treatment of hematopoietic diseases are included.
Abstract:Human hematopoietic stem cells are provided by separation of the stem cells from dedicated cells. The stem cells may than be maintained by regeneration in an appropriate growth medium. Means are provided for assaying for the stem cells as to their capability for producing members of each of the hematopoietic lineages.
