Summary:Described herein are a human hematopoietic stem cell culture and co-culture, methods and uses thereof. The invention relates to a composition of human hematopoietic stem cells comprised of stromal cells, and processes for their isolation and maintenance. Further disclosed are methods of preparing the culture with Thy-1+ and CD34+ human stem cells which are capable of self-regeneration and of differentiation into members of the lymphoid, erythroid and myelomonocytic lineages. Also provided are methods for assaying the stem cells for their capacity to produce members of each of the hematopoietic lineages. Therapeutic applications are included in the regeneration of a deficient hematopoietic system in a host such as after chemotherapy.
Abstract:Human hematopoietic stem cells are provided by separation of the stem cells from dedicated cells. The stem cells may than be maintained by regeneration in an appropriate growth medium. Means are provided for assaying for the stem cells as to their capability for producing members of each of the hematopoietic lineages.
