Summary:Described herein is a method of transferring a nucleic acid into bone progenitor cells using a bone-compatible matrix. Methods and materials are disclosed for the in situ transfer of nucleic acids into bone, such that an osteotropic gene is transferred into bone progenitor cells, which results in the stimulation of the progenitor cells to promote bone growth, repair and regeneration in vivo. Therapeutic applications are included for the treatment of various bone-related diseases and defects, such as in the promotion of fracture repair, for use in conjunction with implants, and in the treatment of osteoporosis and osteogenesis imperfecta.
