Hoshimaru, Minoru; Shiga-ken, Japan | Summary: | Described herein is a retroviral vector containing a regulatable transactivator element and a heterologous gene, and methods of its use in the genetic modification of cells. The invention relates to a novel regulatable retroviral vector in which the v-myc oncogene is driven by a tetracycline-controlled transactivator and a human cytomegalovirus minimal promoter fused to tet operator sequences. Further disclosed are methods by which such a vector may be used for the immortalization of adult neuronal progenitor cells and for the regulation of heterologous Producer cell lines which produce high titers of the recombinant retrovirus.
