Summary:This invention describes a method of introducing large DNA molecules into nonhuman mammals with embryonic stem cells and yeast artificial chromosomes containing foreign DNA. Described herein are embryonic stem cells that are essentially free of yeast DNA and which are prepared from suitably marked yeast artificial chromosomes. Further disclosed are properties of associated vectors, and methods of transforming such an embryonic stem cell by protoplast fusion. Therapeutic and experimental applications in the control of gene expression are included.
Abstract:Embryonic stem cells that are essentially free of yeast DNA are prepared from suitably marked yeast artificial chromosomes and used to transfer DNA segments of considerable size into organisms.
