Summary:Described herein are methods for the ex vivo expansion of multipotent hematopoietic cells according to a process of culturing the cells with a combination of cytokines such as interleukin-6, soluble interleukin-6 receptor and stem cell factor to induce the proliferation and differentiation of hematopoietic stem cells, from which erythroid cell may be produced. Further disclosed are methods by which stem cell factor in combination with gp130 signaling supports such proliferation, differentiation and terminal maturation of erythroid cells from normal human hematopoietic stem cells in the absence of erythropoietin. Therapeutic and experimental applications are provided.
