Summary:Described herein is a population of thymocyte progenitor cells, methods and uses thereof. The invention relates to a subpopulation in the CD4+ 8+ (DP) thymic blast population which is the precursor for thymic T cells, and in which all such progenitors are c-kit+. Further disclosed are properties of the c-kit+ subset which expresses lower levels of CD4 and CD8 than the large and small DP c-kit- cells, such that the DPint c-kit+ cells differentiate to thymic T cells rapidly upon contact with heterogenous thymic stromal cell cultures. Claims of the invention include an in vivo maturation period of 4 days. Therapeutic and experimental applications of such progenitor cells as a source of mature CD4+ and CD8+T cells are included.
