Summary:Described herein is a thrombopoietin polypeptide and ligand for the mpl cytokine receptor, and methods of its detection. The invention relates to techniques for determining the presence or absence of mpl ligand nucleic acids via the hybridization of an oligonucleotide probe and the amplification of a nucleic acid test sample comprised of a nucleic acid polymerase chain that reacts with the oligonucleotide. Further disclosed are recombinant methods by which the mpl ligand is prepared. Claims of the invention include the capacity of the mpl ligands to influence the replication, differentiation and maturation of blood cells, especially megakaryocyte progenitor cells. Therapeutic applications are directed toward the treatment of thrombocytopenia and related diseases.
Abstract:Isolated mpl ligand, isolated DNA encoding mpl ligand, and recombinant methods of preparing mpl ligand are disclosed. These mpl ligands are shown to influence the replication, differentiation or maturation of blood cells, especially megakaryocyte progenitor cells. Accordingly, these compounds are used for treatment of thrombocytopenia.
