Summary:This invention introduces a Streptococcus pneumoniae MurD biosynthetic gene which hybridizes under selective hybridization conditions to a nucleic acid encoding for an enzymatically active protein fragment, namely, uridine-diphosphate-N-acetylmuramyl-L-alanyl-D-isoglutamate ligase. Further disclosed are associated vectors and transformed heterologous host cells for expressing the MurD enzyme product, as are methods and conditions of hybridization. Also provided are techniques for the use of such an enzyme product in the identification of compounds that inhibit stem peptide biosynthesis, such as in the therapeutic detection of antibacterial compounds. This invention represents a variation of other applications filed by the same assignee.
Abstract:The invention provides isolated nucleic acid compounds encoding the murD stem peptide biosynthetic gene of Streptococcus pneumoniae. Also provided are vectors and transformed heterologous host cells for expressing the MurD enzyme product and a method for identifying compounds that inhibit stem peptide biosynthesis.
