, MN, USA
Summary:This invention introduces long-term methods of culturing hematopoietic stem cells or committed progenitor cells in the absence of stromal cells, wherein the conditioned medium comprises an effective added amount of one or more cytokines selected from the group consisting of MIP-1?a, IL-3, SCF, BB10010 and PF-4. Claimed advantages of the invention include an ability to self-replicate and to differentiate which is retained by the hematopoietic stem and committed progenitor cells.
Abstract:A method for the culture of hematopoietic cells is provided comprising culturing said cells in stromal conditioned medium comprising one or more cytokines selected from the group consisting of MIP-1.alpha., IL-3, SCF, BB10010 and PF-4, so that the ability of the stem cells to self-replicate and differentiate is maintained or the ability of the committed progenitors to expand and differentiate is maintained.
