Title: | Methods for gene transfer | Inventors: | Uchida, Nobuko; Palo Alto, CA, USA
Tsukamoto, Ann; Portola Valley, CA, USA
Weissman, Irving; Redwood City, CA, USA | Summary: | This invention provides novel methods of generating transduced hematopoietic stem cells (HSCs) in a mammal. Described herein is a process for the optimization of gene transfer into HSCs following treatment with hydroxyurea prior to gene transfer, which thereby induces the HSCs into the active cell cycle. Therapeutic applications are included in gene therapy and for enhancing the efficacy of bone marrow transplantation by the administration of hydroxyurea to increase HSC yield. | Abstract: | The present invention provides a method for optimizing gene transfer into hematopoietic stem cells (HSCs) by contacting the cells with hydroxyurea prior to gene transfer to induce HSCs in G0 into active cell cycle. This method is useful for treating patients suffering from a disease that is suitably treated by gene therapy or involves hematopoietic cells. A method is also provided for enhancing the efficacy of bone marrow transplantation by administering hydroxyurea to increase HSC yields. | US Patent Website: | Click Here for Full Text of Patent | Title Number: | US5928638 | Application Number: | US1996000664358 | Date Filed: | 17/06/1996 | Date Published: | 27/07/1999 | Assignee: | SyStemix, Inc., Palo Alto, CA, USA |
