Summary:This invention introduces methods for the use of a hepatocyte growth factor to stimulate the proliferation and differentiation of hematopoietic cells. Described herein are novel methods of producing burst-forming unit-erythroid (BFU-E) cells in vitro via a process of treating the cells with an effective amount of a hepatocyte growth factor (HGF). Further disclosed are methods by which the HGF may be obtained from cells transformed with human gene sequences encoding the factor. Pharmaceutical compositions and acceptable carriers are provided, as are therapeutic applications in the treatment of primary or secondary cytopenia and for the ex vivo expansion of bone marrow hematopoietic precursors for transplantation.
Abstract:Hepatocyte growth factor stimulates proliferation and differentiation of hematopoietic cells, most preferably burst-forming unit-erythroid cells. The hepatocyte growth factor may be obtained from cells transformed with human gene sequences coding therefor. Pharmaceutical compositions useful to induce the proliferation and differentiation of hematopoietic cells may contain hepatocyte growth factor as an active principle in admixture with a suitable carrier. The pharmaceutical compositions may additionally contain stem cell factor as an active principle.
