Summary:This invention introduces transgenic nonhuman mammals which are homozygotic for the hepsin gene. Described herein are methods of producing a transgenic mouse comprised of a disrupted hepsin gene via a process of utilizing a hepsin targeting vector for homologous recombination in mouse embryonic stem cells. Claims of the invention include a mouse which exhibits elevated blood serum alkaline phosphatase levels when compared to levels of wild-type mice. Specific nucleotide and amino acid hepsin sequences are provided, as are therapeutic and experimental applications of such an animal model in the treatment of bone diseases.
Abstract:The present invention provides a transgenic mouse comprising a disrupted hepsin gene. In particular, the invention provides methods of making the transgenic mouse comprising the disrupted hepsin gene by utilizing a hepsin targeting vector for homologous recombination in mouse embryonic stem cells. Also, nucleotide and amino acid hepsin sequences are disclosed.
