The invention relates to natural suppressor (NS) cells which secrete a soluble protein suppressor factor (SF) which in turn suppresses the mixed lymphocyte response. Specifically disclosed herein are NS cells with the phenotypes IL-2R+, CD3-, CD4-, CD8-, TCR?a?-, Ig-, and MAC-1-, or which are double negative suppressors (DNS) and have the phenotypes IL-2R+, CD3+, CD4-, CD8-, and TCR?a?+. Claims of the invention include uses of both NS and SF in vivo to confer immunotolerance with respect to allogeneic transplants, to effect immunosuppression, and to enhance engraftment of transplanted cells. Also provided are density gradient separation techniques for enriching progenitor cells as identified by the presence of CD34 surface markers.
