Summary:Described herein is an immunological reagent specific for P-glyco:protein, methods and uses thereof. The invention relates to immunological reagents and methods specific for a mammalian transmembrane protein, Pgp, having a non-specific efflux pump activity which has been identified as a component of multidrug resistance in cancer patients undergoing chemotherapy. Further disclosed are methods for developing and using immunological reagents specific for certain mutant forms of Pgp and for wild-type Pgp in a conformation associated with substrate binding or in the presence of ATP-depleting agents. Claims of the invention include improved methods for purifying hematopoietic stems cells expressing Pgp. Therapeutic applications include diagnostic evaluations of cancer cells expressing Pgp, as well as uses in the detection of multi-drug resistant cancer, in the isolation of hematopoietic cells and in the selective killing of specific cells.
