Summary:This invention describes methods for producing or obtaining gene targeting constructs via homologous recombination in host cells, as well as the resulting targeting constructs per se. The invention also describes transgenic animals having targeted mutations introduced in to the cells of the animal according to this method. Further disclosed are means of preparing shuttle vectors for the transformation of yeast cells and homologous recombination, and isolation of the targeting vectors from this process.
Abstract:The present invention is directed to methods for producing or obtaining gene targeting constructs by way of homologous recombination in host cells and to targeting constructs produced by those methods. The invention is also directed to transgenic animals having targeted mutations introduced in to the cells of the animal using the targeting constructs of the invention.
