Summary:Described herein are novel methods of producing a bovine animal. The invention differs somewhat from the standard somatic cell nuclear transfer (SCNT) process, also known as cloning. Rather than inserting an adult cell into an enucleated oocyte in order to establish a cloned embryo, methods of this invention involve the transfer of “loose suspensions” into an enucleated bovine oocyte, where such suspensions consist of cells from an inner cell mass that has been derived from a normal bovine embryo. The new cloned embryo is then cultured from this oocyte and implanted into a surrogate bovine host where it is allowed to develop into a bovine animal. Claims of the invention include experimental applications in genetic manipulation techniques.
Abstract:The use of totipotent embryonic stem cells to provide substantially identical cells for embryo cloning techniques is described. The method includes the culture of loose suspensions of inner cell mass cells of bovine animals to retrieve large populations of stem cells. The invention also describes the use of stem cells in various genetic manipulation techniques.
