Summary:This invention introduces a method of determining the degree of maturity of stem cells according to a procedure for releasing stem cells from a quiescent state by blocking the cell cycle inhibitor. Described herein is a method for the transfer of a gene into a culture of primitive stem cells which comprises a prestimulation period that is time-limited to less than 36 hours in order to retain hematopoietic potential, and which involves the additional step of adding a blocking agent to block at least one inhibitor of a cell cycle of the primitive stem cells. Claims of the invention include a more efficient transfer and more stable expression, with therapeutic applications in the treatment of genetic diseases.
Abstract:A method is disclosed for gene transfer into a culture of primitive stem cells which comprises a prestimulation step of adding a blocking agent to block at least one inhibitor of a cell cycle of said primitive stem cells. The prestimulation is time-limited for a period of less than approximately 36 hours so that said culture of primitive stem cells retains hematopoietic potential.
