Bishop, Michael D.; Rio, WI, USA
Summary:This invention describes materials and methods for cloning porcine animals. Detailed herein are totipotent cells useful for cloning porcine animals, porcine embryos produced from such cells by employing nuclear transfer techniques, and porcine animals that arise from such cells and such embryos. Fully disclosed are methods of obtaining a totipotent porcine nuclear donor cell from a cell culture, and forming a cybrid by translocating the totipotent porcine nuclear donor cell or nucleus into an enucleated porcine oocyte. Also included are methods by which, prior to enucleation, the oocyte has been matured for between 42 and 56 hours. Details are given for the culturing, translocation, maturation and activation of the oocyte, and for the development of the cybrid into a porcine embryo.
Abstract:The present invention relates to materials and methods for cloning porcine animals. The invention relates in part to totipotent cells useful for cloning porcine animals, porcine embryos produced from such cells by employing nuclear transfer techniques, and porcine animals that arise from such cells and embryos.
