Summary:This invention introduces a novel nucleic acid encoding mammalian protein kinase A anchoring protein (AKAP) 110, methods and uses thereof. Properties of the protein include hybridization with a nucleic acid probe that is useful for the isolation and detection of AKAP110 genes or mammalian gene expression. Further disclosed are recombinant techniques by which cell transformation and homogenous preparations are prepared from the nucleic acid encoding sperm-specific protein kinase A anchoring proteins. Methods of isolation and analytical tools are provided, such as polyclonal antisera and monoclonal antibodies specific for the sperm-specific protein kinase A anchoring proteins, as well as high throughput screening methods for screening compounds that can disrupt binding between protein kinase A and the sperm-specific protein kinase A anchoring protein. Claims of the invention include methods for determining and producing compounds, particularly antisense oligonucleotides, for inhibiting or preventing the expression of sperm-specific protein kinase A anchoring protein in sperm, spermatids, or progenitor cells thereof. Applications are provided in the development of male-specific contraceptives.
