Summary:Described herein are methods and materials for producing a transfusable, oxygenating composition of red blood cells. The invention relates to a process of ex vivo culturing, expansion and differentiation of human primitive hematopoietic cells first in a bioreactor containing one or more growth factors, from which the cells are then differentiated into erythroid progenitor cells in a second bioreactor containing one or more differentiation factors. Further described are the means by which the erythroid progenitor cells are allowed to mature into erythrocytes in a third bioreactor containing one or more maturation factors. Methods are provided for the expansion and differentiation of the transfusable, oxygenating composition of red blood cells, utilizing a bioreactor containing a nutrient medium which includes perfluorocarbons which are described as improving the exchange of oxygen and carbon dioxide between the cells and the nutrient media. Applications in the production of universally compatible red blood cells are included.
