The invention relates to a process of ex vivo culturing, expansion and differentiation of human primitive hematopoietic cells first in a bioreactor containing one or more growth factors, from which the cells are then differentiated into erythroid progenitor cells in a second bioreactor containing one or more differentiation factors. Further described are the means by which the erythroid progenitor cells are allowed to mature into erythrocytes in a third bioreactor containing one or more maturation factors. Methods are provided for the expansion and differentiation of the transfusable, oxygenating composition of red blood cells, utilizing a bioreactor containing a nutrient medium which includes perfluorocarbons which are described as improving the exchange of oxygen and carbon dioxide between the cells and the nutrient media.
