Summary:This invention describes methods of isolation, enrichment and selection of neural cells and neurosphere initiating cells. The invention relates to enriched neural stem and progenitor cell populations, and methods for identifying, isolating and enriching such neural stem cells using a reagent that binds to cell surface markers. Further disclosed are methods for producing a population enriched for human central nervous system stem cells (CNS-SC) which can initiate neurospheres (NS-IC), via a process of selection from a population containing neural or neural-derived cells for cells that bind to the monoclonal antibody AC133 or to the monoclonal antibody 5E12, in order to produce a population enriched for CNS-SC. Therapeutic applications in the treatment of central nervous system disorders are provided.
