Summary:This invention introduces an in vitro process for providing a recombinant, heterologous gene in a genome of a mammalian cell, via the specific replacement of a gene copy by targeted insertion of the heterologous gene, carried on a vector having homologous flanking sequences which reconstruct the complete gene. The method relates to a process for providing a recombinant, heterologous gene in the genome of a eukaryotic cell. In particular, heterologous DNA is inserted into a recipient gene of the eukaryotic cell by homologous recombination. Further disclosed are transgenic or chimeric animals comprising cells with DNA inserted into their genomes by homologous recombination. Additionally, the authors offer instruction in the methods by which these transgenic animal are generated.
Abstract:A process for providing a recombinant, heterologous gene in the genome of a eukaryotic cell is provided. In particular, heterologous DNA is inserted into a recipient gene of the eukaryotic cell by homologous recombination. Moreover, a transgenic or chimeric animal comprising cells with DNA inserted into its genome by homologous recombination is disclosed. Further, the method of making these transgenic animal is taught.
