Summary:Herein are described in vitro methods of producing lymphoid tissue-specific cells, by introducing hematopoietic progenitor and lymphoreticular stromal cells into a porous solid matrix. The invention relates to a method for lymphoid tissue-specific cell production from hematopoietic progenitor cells in a novel three-dimensional culture device, in the presence of lymphoreticular stromal cells and in the absence of exogenously added growth factors which result in the differentiated progeny. Further described are the means by which lymphoid tissue-specific cells may be isolated at any sequential stage of differentiation and further expanded, or genetically altered at any stage of the process. Therapeutic applications are included, such as in transplantation and implantation techniques for the treatment of autoimmune or infectious diseases.
