The invention relates to a method of culturing primordial germ cells and feeder cells in a medium containing factors that elevate cellular cAMP levels. Further disclosed are methods by which such primordial germ cells are isolated from human embryonic tissue, such as from the gonadal ridges of the human embryo, and cultured such that the resulting cells resemble embryonic stem cells or embryonic germ cells in morphology and pluripotency. Also provided are means by which the cells are then maintained several months in culture and can be genetically manipulated using transgenic technology to insert heterologous genetic material.
