Title: | Conditioned media for propagating human pluripotent stem cells | Inventors: | Xu, Chunhui; Cupertino, CA, USA
Gold, Joseph D.; San Francisco, CA, USA | Summary: | Described herein are methods for proliferating human blastocyst-derived pluripotent stem cells in a substantially undifferentiated form, via culturing the stem cells in a growth environment that is essentially free of feeder cells, but which contains an extracellular matrix and a conditioned medium. The invention relates to media that support the growth of primate pluripotent stem cells in feeder-free culture, and cell lines useful for producing such media. Rather than growing pluripotent embryonic cells on feeder layers of primary embryonic fibroblasts, which is a common procedure for preventing the cells from differentiating, this invention describes a process by which standard culture media conditioned by special cell lines can be used to support the proliferation of pluripotent stem cells while inhibiting differentiation in an environment free of feeder cells. Further disclosed are mesenchymal and fibroblast-like cell lines obtained from embryonic tissue or differentiated from embryonic stem cells, and methods for deriving such cell lines, processing the media, and growing the stem cells using the conditioned media. | Abstract: | This invention provides media that support the growth of primate pluripotent stem cells in feeder-free culture, and cell lines useful for producing such media and other purposes. Conventionally, it has been necessary to grow pluripotent embryonic cells on feeder layers of primary embryonic fibroblasts, in order to prevent them from differentiating. It has now been discovered that standard culture media conditioned by special cell lines can be used to support proliferation of pluripotent stem cells while inhibiting differentiation in an environment free of feeder cells. This invention includes mesenchymal and fibroblast-like cell lines obtained from embryonic tissue or differentiated from embryonic stem cells. Methods for deriving such cell lines, processing media, and growing stem cells using the conditioned media are described and illustrated in this disclosure. | US Patent Website: | Click Here for Full Text of Patent | Title Number: | US6642048 | Application Number: | US2001000900752 | Date Filed: | 06/07/2001 | Date Published: | 04/11/2003 | Assignee: | Geron Corporation, Menlo Park, CA, USA |
