Stem Cell Related Patent Number US6723561
Title: | Materials and methods relating to the transfer of nucleic acid into quiescent cells | Inventors: | Russell, Stephen James; Cambridge, United Kingdom
Fielding, Adele Kay; Cambridge, United Kingdom
Casimir, Colin Maurice; London, United Kingdom | Summary: | Described herein is a novel retroviral packaging cell line composed of new retroviral particles and a nucleic acid encoding a therapeutic polypeptide which is incorporated into quiescent cells. The invention relates to methods and materials for transferring the nucleic acid encoding a polypeptide for treating a disease or disorder into populations of quiescent cells such as hematopoietic stem cells (HSCs). Further disclosed are retroviral packaging cell lines and retroviral particles expressing and displaying a growth factor such as stem cell factor (SCF) on the cell surface, or a fusion with a viral envelope protein. Also described are methods and compositions comprising the retroviral packaging cell lines and retroviral particles, and their in vivo and ex vivo uses. Therapeutic applications in the treatment of a variety of diseases are included. | Abstract: | Materials and methods for transferring nucleic acid encoding a polypeptide for treating a disease or disorder into populations of quiescent cells such as haematopoietic stem cells (HSCs), using retroviral packaging cell lines and retroviral particles expressing and display a growth factor such as stem cell factor (SCF) on the cell surface or as a fusion with a viral envelope protein. The present invention also relates to compositions comprising the retroviral packaging cell lines and retroviral particles, and their use in methods of medical treatment, in vivo and ex vivo. | US Patent Website: | Click Here for Full Text of Patent | Title Number: | US6723561 | Application Number: | US1998000043665 | Date Filed: | 05/10/1998 | Date Published: | 20/04/2004 | Assignee: | Mayo Foundation for Medical Education and Research, Rochester, MN, USA |
