Summary:Described herein is a novel retroviral packaging cell line composed of new retroviral particles and a nucleic acid encoding a therapeutic polypeptide which is incorporated into quiescent cells. The invention relates to methods and materials for transferring the nucleic acid encoding a polypeptide for treating a disease or disorder into populations of quiescent cells such as hematopoietic stem cells (HSCs). Further disclosed are retroviral packaging cell lines and retroviral particles expressing and displaying a growth factor such as stem cell factor (SCF) on the cell surface, or a fusion with a viral envelope protein. Also described are methods and compositions comprising the retroviral packaging cell lines and retroviral particles, and their in vivo and ex vivo uses. Therapeutic applications in the treatment of a variety of diseases are included.

The present invention also relates to compositions comprising the retroviral packaging cell lines and retroviral particles, and their use in methods of medical treatment, in vivo and ex vivo.
