Title:Method of producing mutant mice

Summary:This invention describes a method of producing a mutant targeted non-human mammal by introducing pluripotent cells into tetraploid blastocysts of the same mammalian species and transferring the resultant embryo into a foster mother, from which live offspring develop. Such a method does not require the production of chimera and permits the introduction of multiple mutations in the embryos, thereby avoiding the necessity of breeding to combine all of the desired mutations in a single animal. Further disclosed are applications of this method to the identification of drugs, and various ways in which this invention represents improvements over previous methods of producing mutant mice.
Abstract:A method of producing mutant/targeted non-human mammals, such as mutant mice that does not require production of chimera and permits the introduction of multiple mutations in embryos and, thus, avoids the necessity of breeding to combine all of the desired mutations in a single animal. The method is efficient in producing ES mice.
