Summary:Described herein are novel purified nucleic acid molecules comprising cardiac-specific cell enhancer elements derived from Csx/Nkx2.5 regulatory regions. Further disclosed are the uses of such enhancer elements in the regulation of gene expression in cardiac cells, in the process of inducing stem cells (e.g., embryonic stem cells or bone marrow stem cells) to differentiate as cardiomyocytes, and in the identification of factors that induce the differentiation of stem cells as cardiomyocytes. Therapeutic applications are included, such as in the expression of genes in cardiac cells, as earlier markers of cardiomyocyte induction, and for the optimization of cardiomyocyte induction.
Abstract:The invention features cardiac cell-specific enhancer elements derived from Csx/Nkx2.5 regulatory regions. These enhancer elements are useful, for example, for (i) regulating gene expression in cardiac cells, (ii) inducing stem cells (e.g., embryonic stem cells or bone marrow stem cells) to differentiate as cardiomyocytes, and (iii) identifying factors that induce the differentiation of stem cells as cardiomyocytes.
