Stem Cell Related Patent Number US6828145

Title:Method for the isolation of stem cells by immuno-labeling with HLA/MHC gene product marker

Summary:Described herein is a method of isolating stem cells of a mammal by isolating those which do not express ?2 -microglobulin. Central to the invention is the discovery that mammalian stem cells do not express ?2 -microglobulin, allowing the method disclosed herein to capitalize upon a process of sorting and separating, from a sample of cells, those cells that express ?2 -microglobulin from those cells that do not express ?2 -microglobulin. Further described are the means by which stem cells are sorted and selected from the population of cells accordingly. Also disclosed is the known identifiable marker which some cells in this process are found to express, such as proteins expressed by genes encoding the major histocompatibility complex. Methods of identification and separation are provided.
Abstract:Disclosed herein is the discovery that mammalian stem cells do not express .beta..sub.2 microglobulin (.beta..sub.2 m). The invention discloses a method of isolating stem cells comprising sorting, from a sample of cells, cells that express .beta..sub.2 m from cells that do not express .beta..sub.2 m. One then selects stem cells from the population of cells that does not express .beta..sub.2 m. This is accomplished by selecting cells that express a known marker, such as proteins expressed by genes encoding the major histocompatibility complex. An isolated stem cell is disclosed, as is a method for identifying it and other stem cells.
