The invention relates to antibodies that have binding specificity for an antigen that is expressed on a subset of human, hematopoietic mononuclear cells, including a hematopoietic stem cell population, but is not expressed on normal, mature myeloid cells. Disclosed herein are properties of a polynucleotide molecule encoding a polypeptide, a fragment or variant thereof, which is expressed on the surface of a subset of human hematopoietic mononuclear cells, such that the polypeptide specifically immunoreacts with the MG1 antibody. The monoclonal antibody, MG1, is described as useful in methods of isolating cell suspensions from human blood and marrow that can be employed in bone marrow transplantation, genetic therapy, and in the treatment of other diseases of the hematopoietic system. Further disclosed are cell suspensions containing MG1<+> human hematopoietic cells, as well as therapeutic methods employing the cell suspensions. Specific amino acid sequences and properties of the novel antigen are provided. Also included are therapeutic applications in the diagnosis and treatment of leukemia, for bone marrow reconstitution and in gene therapy. | Abstract: | The subject invention pertains to antibodies that have binding specificity for an antigen that is expressed on a subset of human, hematopoietic mononuclear cells, including a hematopoietic stem cell population, but is not expressed on normal, mature myeloid cells. In one embodiment, a monoclonal antibody, MG1, is provided. This antibody is useful in methods of isolating cell suspensions from human blood and marrow that can be employed in bone marrow transplantation, genetic therapy, and in treating other diseases of the hematopoietic system. Cell suspensions containing MG1+ human hematopoietic cells are also provided, as well as therapeutic methods employing the cell suspensions.
