Summary:This invention describes a method of engineering an improved organism with a desirable trait, generated from an initial population of organisms, and which in turn generates a set of mutagenized organisms. The invention relates to the cellular transformation, directed evolution, and screening methods for the creation of novel transgenic organisms with specifically desired properties. Described herein are methods of generating a transgenic organism such as a microbe or a plant, having a plurality of traits that are differentially activatable, as well as methods of reprogramming genes and gene pathways by the introduction of regulatory sequences, such as promoters, that are operable in an intended host, and which thereby confer operability to a novel gene pathway when it is introduced into an intended host. Therapeutic applications are included.
