Summary:This invention describes a new transgenic nonhuman mammal, comprising two human immunoglobulin loci. Specifically, described is a transgenic mouse wherein one of the human immunoglobulin loci is a human heavy chain locus, and the other locus is a human light chain locus, such that the human heavy chain locus is carried by transchromosome SC20 and the human light chain locus is carried by a transgene integrated into the genome of the mouse. Additionally, an endogenous mouse heavy chain locus and at least one endogenous mouse light chain locus are inactivated. Disclosed herein are the mechanisms by which such a novel transgenic mouse is capable of producing human sequence antibodies, and also discussed are various uses of these antibodies, such as in the generation of B cells expressing human sequence antibodies, and in the generation of antigen-specific hybridomas secreting human sequence antibodies.
Abstract:The present invention provides novel transgenic nonhuman mammals capable of producing human sequence antibodies, as well as methods of producing and using these antibodies.
