Summary:This invention describes a method of delivering a polynucleotide to a human stem cell by linking the polynucleotide with a human sperm cell through a linker and by fertilization of the human oocyte, resulting in a zygote that is cultured and established as human embryonic stem cells. The invention relates to a vector system for delivering a gene into a human stem cell via a monoclonal antibody mAbC, wherein the antibody is characterized by having binding affinity to a sperm cell, such that the sperm cell bound with the antibody retains the ability to fertilize an oocyte, and wherein the antibody is secreted by the hybridoma assigned ATCC accession number PTA-6723. Applications for cell transplantation into the human body are included.
Abstract:A method and vector system for delivering of a gene into a human stem cell for therapeutic uses is disclosed. The method includes linking a therapeutic gene to human sperm cells through a linker and fertilizing a human oocyte. The resulting zygote may then be cultured and established as human embryonic stem cells, which may later be differentiated into different types of cells for transplantation into the human body.
