Title:In vivo screening methods using enriched neural stem cells
Inventors:Buck, David W.; Heathfield, United Kingdom
Uchida, Nobuko; Palo Alto, CA, USA
Weissman, Irving; Redwood City, CA, USA
Summary:Described herein are methods of isolation, enrichment and selection of neural cells and neurosphere-initiating cells (NS-IC). Also disclosed are enriched neural stem and progenitor cell populations, and methods for the identification, isolation and enrichment of such cells using a reagent that bind to cell surface markers. The invention relates to a method of drug screening which incorporates selecting from a population of neural or neural-derived cells those cells which bind to the monoclonal antibodies AC133 or 5E12, thereby producing a population enriched for human NS-IC. Also provided are methods of engrafting, administration and comparison. Applications in the treatment of central nervous system disorders are included.
Abstract:Enriched neural stem and progenitor cell populations, and methods for identifying, isolating and enriching for neural stem cells using reagent that bind to cell surface markers, are provided.
