Summary: | Described herein are methods of culturing undifferentiated embryonic stem (ES) cells via culturing of the ES cells in the presence of BMP-mediated default pathway antagonists. This invention provides a preparation of undifferentiated ES cells sustainable for a prolonged period in an undifferentiated state which will undergo stem cell renewal or somatic differentiation. Further disclosed are means by which the cells are capable of somatic differentiation in vitro and are inclined to differentiate away from an extra-embryonic lineage. Also provided are methods of culturing ES cells to improve stem cell maintenance and persistence in culture. The techniques disclosed herein include a culture of ES cells as well as differentiated cells derived from the embryonic cells that result from directed differentiation procedures. |
