Title:Use of Xenopus laevis oocytes a microincubators
Inventors:Paylian, Sergei; Knoxville, TN, USA
Summary:Herein are described methods for the de-differentiation and stabilization of cells, such as progenitor cells, by introducing the cells into the cytoplasm of host oocytes, such as those of Xenopus laevis. The authors describe benefits of this method, which include obviation of the need for any nuclear transfer procedure, which is known to disrupt the chromosomal architecture of donor and recipient cells. Further described is a microincubator comprising an isolated host Xenopus laevis oocyte having at least one bone marrow stromal (BMS) cell encapsulated within the cytoplasm of the host oocyte. Cells that have been removed from the host oocytes are also addressed, as are the means by which the host oocytes encapsulating the cells have been introduced into their cytoplasm.
Abstract:The present invention concerns methods for de-differentiation and stabilize cells, such as progenitor cells, by introducing the cells into the cytoplasm of host oocytes, such as Xenopus laevis oocytes. Advantageously, this method obviates the need for any nuclear transfer procedure, which is known to disrupt the chromosomal architecture of donor and recipient cells. The present invention also concerns host oocytes encapsulating cells that have been introduced into their cytoplasm. The present invention also concerns cells that have been removed from the host oocytes.
