Summary:This patent is a reissue of patent no. US5830701. Described herein is a method of counting hematopoietic progenitor cells using flow cytometry without the use of antibodies. Methods are described for the detection of immature cells, the construction of a cell distribution profile and the detection of sub-classes of hematopoietic progenitor cells. The invention relates to improved methods for detecting and/or counting the appearance of hematopoietic progenitor cells, which consists of a novel protocol for mixing a blood sample with a reagent that is capable of detecting immature cells, without employing immunological techniques. Information is gathered on the treated sample via the use of a particle analyzer and the constructed cell distribution profile, a delineated portion of which is characterized as a zone in which at least one subclass of hematopoietic progenitor cells appear, and the cells within that zone are counted. The claims of the invention include an improvement in simplicity and accuracy over previous detection methods.
