Described herein is a method of counting hematopoietic progenitor cells using flow cytometry without the use of antibodies. Methods are described for the detection of immature cells, the construction of a cell distribution profile and the detection of sub-classes of hematopoietic progenitor cells. The invention relates to improved methods for detecting and/or counting the appearance of hematopoietic progenitor cells, which consists of a novel protocol for mixing a blood sample with a reagent that is capable of detecting immature cells, without employing immunological techniques. Information is gathered on the treated sample via the use of a particle analyzer and the constructed cell distribution profile, a delineated portion of which is characterized as a zone in which at least one subclass of hematopoietic progenitor cells appear, and the cells within that zone are counted.
